I've done my best going down the lineup, and listening to the songs each artist has provided on the Roo site, but I'm afraid there's a lot I'm not noticing. I went to Lolla in '09 and missed one of my now-favorite bands because someone showed me a song of theirs that I'm still not digging on.
So far I have: Paul, Bjork, PL*, Byrne & Vincent, Passion Pit, Grizzly Bear, AnCo, Beach House*, ZZ Top*, Cat Power, Tame Impala (checking them out for the first time right now, I dig), Boys Noize, Bush and McCoury, Matt & Kim*, Alt-J, John McLaughlin, Father John Misty, A-Trak, Preservation Hall*, Earl Sweatshirt, Paper Diamond* (saw at Ultra, not awesome), Purity Ring, Swans, Araab, Cults, Four Tet, Death Grips, Wild Nothing, DjangoDjango, SeaWolf.

Two must sees for me are Local Natives and Milo Greene.
I think you have too many on your list, though. I'd be impressed if you were able to see 30 sets, not to mention to inevitable scheduling conflicts these acts above will most likely face.
Preservation hall is always always a treat, I see them every time I go to the jazzfest in NoLA. Frank Turner is great folky rocky way check him out some more if you got time. Matthew e whites great also he has a chill sound on this album he is touring I can't get enough of it. Dwight Yoakum is a country legend. If you are remotely interested in or just plain curious about country music this is really a great show. Calexico has a bitchin' western movie sound going for them. Lee fields has soul to spare. Wilco is well wilco alt-country mellow psychedelia
This is a really deep lineup. I hope to catch 30 sets but I will be happy with five must see shows a day.
Keeping headliners out of it, since they're unopposed, my only "will not miss for anything" for me is the Soul Superjam. You probably can't go wrong catching Jim James' solo set either. If you like Americana at all, Jason Isbell puts on a solid show.

Listened to Lord Huron the other dayfor the first time and theyve turned into a can't miss for me...I've Local Natives for a while now and they're another can't miss....Lord Huron and Local Natives are can't miss indie bands for me since I've only heard great things about their live shows.
